what exactly do you mean by isolate?

if you're trying to pick out parts from a whole mix, maybe a surgical eq can help.  try A0 parametric EQ (but i doubt if it's surgical enough).  it's really difficult to isolate one instrument from the other once the song has been mixed.  there are soime parts that are in the same frequency range and chances are these parts will also disappear if you EQ them out.

well if you want to remove a lead vocal from a stereo mix, it may be possible
if its a vocal that is panned dead center. but the main conditions are if:
1. lead vocal only on a stereo mix
2. not too much reverb/digital effects  (1960 to early 1980's stuff)

results will vary, but sometimes you get to eliminate the vocal but still
retain a sizable portion of the instruments.

sound forge has its channel converter (remove center material from stereo mix) and most audio editors will have it as a dx/vst plug in

now if you want to isolate guitar solos or bass runs, thats another matter,
however there's no harm in trying...

try eq'uing out everything  from 700 hz up.  maybe 250 HZ below will even isolate the bass better.  just use a parametric EQ with a graphical display (low pass engaged) and "sweep" through higher frequencies to see what filters everything out and leaves the bass best. you wont be able to completly isolate just the bass but like he said, it's worth a try.
